Gold for culture level

I am new to the game ans have won some gold in quest, for which I have used 50 gold in 2 cities which I though would give one level however now 100 gold down it hasn`t even given a point let alone a level, can any advise where i`ve gone wrong?

Also is it possible to drop a city to free a slot?

it takes 6 hours for the olympic games (50 gold) to produce a culture point.
